报告摘要:

Transmembrane proteins facilitate the exchange of materials, energy, and information across the cell membrane, thereby enabling cells to adapt more effectively to the surrounding environment. There has been de novo designed transmembrane proteins; however, they lack the ability to respond to specific environmental cues—such as voltage, pH, ligands, mechanical force, light, and temperature. Here, we report the de novo design of voltage-gated ion channels and ligand-binding transmembrane proteins. These designer proteins are functional in mammalian cells upon expression. Cryo-EM and crystal structures of the designed transmembrane proteins show a high agreement with our design models. Collectively, our results contribute novel insights into membrane biology and open avenues for a diverse array of applications.

卢培龙,西湖大学生命科学学院特聘研究员。研究方向为蛋白质设计,进行合成生物学、生物物理学与计算生物学多学科交叉研究。精确设计了多次穿膜蛋白质的三维结构;完成了纳米孔膜蛋白的精确设计,证明从头设计的跨膜蛋白可以被精准设计并行使功能。在Science, Nature等期刊以第一作者或共同通讯作者身份发表多篇论文。主持国家重点研发计划和国家自然科学基金等项目。
